세포 유전체에는 발달 및 분화과정에서 표현형에 영향을 미치는 즉, 유전자 전사에 영향을 미치는 몇가지 알려져 있는 regulatory elements이 존재한다. 여기에는 Promoter, Enhancer, Insulator 영역 등이 존재하고(cis-regulatory elements), 이는 더 나아가 TAD(topologically associated domains)이라는 큰 영역 안에서 직접적 또한 간접적 유기적인 관계로 gene 발현을 조절하는 것으로 알려져 있다. 또한, 이러한 영역들은 gene을 transcript하기 위하여 해당 영역에 transcription factors(TFs)이 영역을 인지하고 함께 binding하여 complex를 이루게 된다. 하지만, TFs가 binding하는 영역에 genetic mutation이 발생하게 된다면, 비정상적인 TFs binding이 일어나게 되고, 즉 motif rewiring이 일어나게 된다. 결론적으로 proto-oncogene을 oncogene으로써 또는 tumor suppressor 역할을 하고 있던 유전자를 억제할 수 있다. 이는 더 나아가 암의 원인(tumorigenesis), 예후(prognosis, drug response) 등에 영향을 미칠 수 있다. 해당 연구는 motif rewiring을 야기시키는 somatic alteration에 집중하여 genetic alterations이 epigenetic alteration에 유기적인 관계를 확인하고, 한국인 대장암의 원인 또는 예후에 영향을 미치는 biomarker 발굴에 주력하는 프로젝트이다. |
